Our Treatments This study is investigating the effects of  BPL-003 (a formulation of the psychedelic 5-MeO-DMT) on Treatment Resistant Depression. Ultra-short-acting psychedelics like dimethyltryptamine (DMT) and 5-MeO-DMT may be advantageous compared to longer-acting psychedelics like psilocybin in terms of practicality for use as therapeutic interventions in clinical settings. 5-MeO-DMT is being developed and evaluated for potential therapeutic effects in patients with treatment-resistant depression (TRD). For example, the potencies of drugs substituting for 5-MeO-DMT in drug discrimination assays is well-correlated with their serotonin 5-HT1A receptor affinities, and the discriminative stimulus effects of 5-MeO-DMT are attenuated by serotonin 5-HT1A receptor antagonists. Introduction and Rationale for Study Other 5-methoxytryptamines like 5-MeO-DET and 5-MeO-DPT were plagued by dose-limiting side effects that resulted in Shulgin having an unfavorable impression of them, whereas 5-MeO-DiPT and 5-MeO-DALT produced only light psychedelic effects. 5-MeO-DMT was one of the only psychedelics in TiHKAL that resulted in a plus-four experience on the Shulgin Rating Scale. With harmaline doses of 70 to 150 mg orally, 5-MeO-DMT becomes orally active at doses of 10 to 25 mg, the onset is 15 to 18 minutes, peak effects occur after 1.5 hours, and the duration is at least 3 or 4 hours. This case study was exempt from ethics review and approval, in line with the Baylor College of Medicine Human Research Protections Manual, including Institutional Review Board procedures. The data presented here were collected by the subject for their own interest and safety, and to monitor their progress over time. Electrophysiological effects of 5-MeO-DMT Under drug-free conditions, state maps usually display very similar patterns across animals, despite inter-individual differences in behaviour and LFP signals. A second non-exclusive hypothesis is that the peripheral binding of psychedelics causes fewer molecules to reach the CNS. Classical psychedelics such as 5-MeO-DMT can also bind to serotonergic receptors of the peripheral nervous system (PNS) and this could affect the CNS quite differently from the ICV dosing. The administration of 5-MeO-DMT induced significant alterations in animal locomotion, which is an important factor to consider when interpreting the potential effects observed in electrophysiology, as speed can have a profound impact.
