Then there’s integrated information theory (IIT), a mathematical and philosophical theory that stands out from the rest because it doesn’t start with the brain. If you have ever seen something that wasn’t there simply because you expected to see it, you know how much our brain’s predictions can govern what we actually perceive. The buzz must also have a corresponding “meta-representation” in the frontal parts of the brain that are responsible for higher-order thinking—such as the thought “I am 5-meo-dmt hearing the refrigerator buzzing.” Further, mRNA for the enzyme necessary for the production of DMT, INMT, are expressed in the human cerebral cortex, choroid plexus, and pineal gland, suggesting an endogenous role in the human brain. DMT is naturally occurring in small amounts in rat brains, human cerebrospinal fluid, and other tissues of humans and other mammals. Several speculative and yet untested hypotheses suggest that endogenous DMT is produced in the human brain and is involved in certain psychological and neurological states. In summary, our findings indicate that 5-MeO-DMT administration leads to a decrease in theta power in the hippocampus for equivalent locomotion speeds, as well as an increase in delta power specifically following IP injections. As the drug significantly affected locomotion (Fig. 1B) and speed is known to influence LFPs, we employed a generalised linear model (GLM) to assess the effect of 5-MeO-DMT administration on delta (1–4 Hz) and theta (5–12 Hz) power while accounting for speed (Fig. 2, see Methods for details). Explore by subject Thus, the 5-HT system is rather well-placed to regulate brain state quality or intensity rather than impose a unidirectional effect of all-or-none state switching, which is consistent with the occurrence of a mixed, dissociated state induced by psychedelics. Thus, we believe that the brain-wide 5-HT circuitry is likely to be involved in the response to psychedelics administered systemically. Further work is necessary to address the underlying circuit mechanisms, which could, for example, implicate cortical layer 5, known to play an important role in the generation of SWA63, and shown to be essential in mediating the effects of certain psychedelics64. On the other hand, the suppression of theta activity and increased pupil dilation were effectively prevented by the injection of a 5-HT1A receptor antagonist. Furthermore, the 5-MeO-DMT-generated slow waves during wakefulness, although not impacted directly by sleep pressure, were followed by lowered SWA during NREM sleep if the drug was administered after sleep deprivation.
